dear bro there is no problem in placing a youtube video in your signature.
All you have to do is paste the direct link [url of video] in your signature and click save signature.

But there is a small condition you can only use video signature if you are active in HG media section.
active means you regularly reply to videos in media section.

or

you have added a minimum of 50 videos in HG Media section these videos could be of any type from islamic to political or entertainment click the link to see all the catagories of videos
